چکیده
Optimum parameters for retention of motility of Treponema pallidum (Nichols virulent strain) were found by anaerobic co-incubation of the treponeme with rat glial cells and anaerobic incubation in spent medium obtained from glial cells originally grown aerobically.

Examination of various cell culture techniques for co-incubation of virulent Treponema pallidum (Nichols I strain) under anaerobic conditions.
Treponema pallidum (Nichols virulent) was incubated with and without cells in cell culture medium reduced to -275 mV Ecal, pH 7.3, under deoxygenated conditions. Five to ten percent of the treponemes attached to cells and remained motile for at least 120 h in cell-treponeme systems of co-incubation. Virulent treponemes could be detected after 120 to 144 h in the supernatant fluids of cell-trepo...
متن کاملProlonged survival of virulent Treponema pallidum (Nichols strain) in cell-free and tissue culture systems.
Survival of Treponema pallidum was found to be prolonged in the presence of tissue culture. Of the 12 cultures studied, cottontail rabbit epithelium (Sf1Ep) supported T. pallidum for the longest time. In horizontal Leighton tubes with reduced medium and an atmosphere of 5% CO2 in N2, the 50% survival time (ST50) was 5 to 6 days for treponemes associated with monolayers of Sf1Ep cells. Comparabl...
متن کاملRedox potential and survival of virulent Treponema pallidum under microaerophilic conditions.
A strongly reduced culture medium, capable of maintaining the virulence of Treponema pallidum (Nichols) for several days, was exposed to an atmosphere of 3% oxygen in nitrogen for 2-3 days before inoculation with T pallidum. By using various volumes of medium in uniform tubes a range of redox potentials (Ecal) from -94 mV to -325 mV was produced depending on the surface area-to-volume ratios of...
متن کاملTreponema pallidum (Nichols strain) in tissue cultures: cellular attachment, entry, and survival.
The interaction of Treponema pallidum (Nichols strain) with cultured cells was investigated under aerobic conditions. Cell monolayers derived from rabbit testicular tissue extended the survival of treponemes as indicated by active motility. Large numbers of organisms rapidly attached to cultured cells. Within 3 h, one to twelve actively motile treponemes were attached to 25 to 50% of the cells....
متن کاملIn vitro culture system to determine MICs and MBCs of antimicrobial agents against Treponema pallidum subsp. pallidum (Nichols strain).
A new procedure for determining the susceptibility of Treponema pallidum subsp. pallidum to antimicrobial agents was developed, utilizing a tissue culture system which promotes the in vitro multiplication of this organism. In the absence of antibiotics, T. pallidum (Nichols virulent strain) multiplied an average of 10-fold when incubated for 7 days in the presence of Sf1Ep cottontail rabbit epi...
